The maiden flight was from Harare International Airport to Buffalo Range Airport in Masvingo, one of the country's key tourism regions on the border with South Africa.
Skylink will link Hwange National Park, Kariba, Masvingo, Mutare and other major tourist resorts on selected days of the week.
The move is aimed at improving the country's airlift systems ahead of the World Cup tournament slated for South Africa.
